What Is Epitalon and Why Is It Also Spelled Epithalon?

Epitalon is a synthetic tetrapeptide, a chain of just four amino acids: alanine, glutamic acid, aspartic acid and glycine. It was developed at the St Petersburg Institute of Bioregulation and Gerontology, in the group led by Professor Vladimir Khavinson, as a small, defined molecule based on epithalamin, a peptide extract the same group had prepared from cattle pineal glands. Most of the published Epitalon bioregulator literature comes from that institute, which describes short peptides of this kind as “peptide bioregulators”.

So is it Epitalon or Epithalon? Both names refer to the same molecule. Epitalon also known as Epithalon is the result of transliteration from Russian (Эпиталон), where the “th” sound is written without an “h”. The longer spelling follows the English word epithalamus, the brain region that contains the pineal gland, and epithalamin, the extract the peptide was modelled on. In searches and on supplier labels, Epitalon vs Epithalon is only a spelling difference; you will also see “Epithalone”. What Does AEDG Mean in Epitalon Research?

AEDG is the peptide’s sequence written in one-letter amino acid codes: A for alanine, E for glutamic acid, D for aspartic acid and G for glycine, read from the N-terminus (the free amino end) to the C-terminus (the free acid end). Many research papers call the molecule “AEDG peptide” rather than Epitalon, so searching both names is the best way to find the full body of work on Epithalon peptides. The same group has studied other short peptides with similar letter codes, which is why papers often refer to the sequence rather than a trade name.

Amino Acid Sequence, Molecular Weight and Chemical Structure

H-Ala-Glu-Asp-Gly-OH (AEDG)

• Molecular formula: C14H22N4O9
• Molecular weight: about 390.35 g/mol (average); monoisotopic mass about 390.139
• CAS number: 307297-39-8
• Structure: a short linear chain with no ring or disulfide bridge. Two of the four side chains (glutamic acid and aspartic acid) are acidic, so with the C-terminal carboxyl group the peptide carries a net negative charge at neutral pH and dissolves readily in water.
• Chemical notes: it contains no methionine, cysteine or tryptophan, the residues most prone to oxidation. It does contain an Asp-Gly bond, which in solution can slowly rearrange into an iso-aspartate form. This is one reason the dry powder is far more stable than a solution.
• Salt form: if supplied as Epitalon acetate, the measured mass on the COA is still that of the peptide itself; the acetate shows up only in net peptide content.

How Is Epitalon Studied in Laboratory Research?

Published Epitalon research falls into three main areas: telomeres and cell ageing in cultured cells, pineal gland and melatonin regulation in animals, and gene expression studies on how such a short peptide might act inside cells. When people ask what does Epithalon peptide do, the honest answer is what it does in those experiments. Most of the work comes from one research group, and the findings below are not evidence of any effect in people.

Telomerase and Cell Ageing Research

Telomeres are protective DNA caps on the ends of chromosomes that shorten each time most cells divide. Telomerase is the enzyme that can rebuild them. In 2003, Khavinson and colleagues reported that adding Epitalon to cultures of human somatic cells switched on telomerase activity and lengthened telomeres (Bull Exp Biol Med, 2003). A follow-up study from the same group reported that treated human fibroblasts kept dividing for more generations than untreated cells [VERIFY: Khavinson et al., Bull Exp Biol Med, 2004].

In 2025, an independent team at Brunel University London tested this again. They reported that Epitalon increased telomere length in normal human epithelial cells and fibroblasts, with higher hTERT gene expression and telomerase activity. In two breast cancer cell lines, telomeres also lengthened, but through a different route called ALT (alternative lengthening of telomeres) (Al-dulaimi et al., Biogerontology, 2025). Because longer telomeres in cancer cells are a concern rather than a benefit, that result is a reason for careful further research, not a selling point.

In animals, a 2003 lifetime study in female mice reported no change in average life span or body weight, a 17% lower rate of chromosome aberrations in bone marrow cells, and a 12% longer maximum life span than in controls (Anisimov et al., Biogerontology, 2003). These Epithalon uses in research are about ageing biology in cells and rodents.

Pineal Gland and Melatonin Research

The pineal gland is a small gland in the brain that makes melatonin, the hormone that tracks the daily light and dark cycle. It sits in the epithalamus, and epithalamus function centres on linking light signals to these daily rhythms. Because Epitalon was modelled on a pineal extract, the Epitalon pineal gland question has been a main research theme, and it is often described as an Epitalon pineal bioregulator.

In a 2001 study in older female rhesus monkeys, Epitalon increased evening melatonin production and brought the daily cortisol rhythm closer to that seen in younger animals (Khavinson, Goncharova and Lapin, Neuro Endocrinol Lett, 2001). Similar effects on melatonin were later reported in older people in small studies by the same group [VERIFY: Korkushko et al., 2007]. These are measurements of hormone levels in research settings, not evidence of any effect on sleep.

What Does Current Evidence Say About Human Studies?

Human Epitalon studies exist, but they are limited. Almost all come from Khavinson’s institute and its partners, and many used epithalamin, the pineal extract, rather than synthetic Epitalon:

• Long-term follow-up of older adults: a 2003 report followed 266 people over 60 for six to eight years after courses of epithalamin and the thymus extract thymalin, and reported lower mortality than in an untreated group (Khavinson and Morozov, Neuro Endocrinol Lett, 2003). The design and reporting do not meet modern standards for a placebo-controlled, blinded trial.
• Smaller clinical reports: studies of hormone rhythms, blood-cell telomere length and other markers in older people have been published by the same group [VERIFY specific papers before naming].
• Independent trials: we found no completed, independently run, placebo-controlled trial of synthetic Epitalon with published results [VERIFY on ClinicalTrials.gov before publishing].

A 2025 review of Epitalon peptide research called the evidence promising but noted that most data come from preclinical work and a single centre (Araj et al., Int J Mol Sci, 2025). Epitalon is not an approved medicine in the UK, the EU or the US, and none of the findings on this page should be read as evidence of safety or effect in humans.

How to Read HPLC Purity and Mass Spectrometry Results

• HPLC purity: the trace shows one main peak for the peptide and smaller peaks for impurities, such as shortened sequences or rearranged forms. Purity is the main peak’s area as a percentage of all peaks. This batch: [CONFIRM: __%].
• Mass spectrometry: for Epithalon peptide the expected singly charged ion [M+H]+ is about 391.1, with a sodium adduct [M+Na]+ sometimes seen at about 413.1. A clear match confirms identity. A peak 18 mass units lower can point to the Asp-Gly ring-closed (succinimide) impurity.
• Net peptide content (if shown): the share of the powder that is peptide, as opposed to counter-ions (usually acetate) and residual water. It is usually below 100% even when purity is high.
• Check the details: the product name, batch number, test date and lab name should all be on the certificate.

How to Store and Handle a Lyophilised Peptide Vial
